Results for Bisphenol A

Increased susceptibility to adult-induced precancerous lesions and hormonal carcinogenesis

Subjects: SD rats

Chemical: Bisphenol A

Low doses tested: 10 μg/kg bw/d

Route of administration: injected subcutaneously

Exposure duration: post-natal day 1 – post-natal day 5 (comparable to human prenatal development from approximately day 1 of week 20 to day 1 of week 26)

Age of measurement: 10, 90 and 200 days of age

Results for Bisphenol A

Down regulation of testicular ERα and ERβ transcript with hypermethylation of CpG sites in the promotor regions of ERα and ERβ. Dnmt3a and Dnmt3b mRNA transcript and protein levels were upregulated in adults.

Subjects: Holtzman rats

Chemical: Bisphenol A

Low doses tested: 2.4 μg/rat

Route of administration: dissolved in sesame oil and delivered subcutaneously

Exposure duration: post-natal day 1 – post-natal day 5 (comparable to human prenatal development from approximately day 1 of week 20 to day 1 of week 26)

Age of measurement: post-natal day 125

Results for Bisphenol A

Increased demethylation of Nsbp1 on post-natal days 10, 90 and 200 with increased methylation of Hpcal1 on post-natal days 10 and 90. On day 10 DNMT1, DNMT3, MBD2, and DNMT3A expression was increased while MBD1 expression was decreased. DNMT3B, MBD2 and MBD3 expression was increased on day 90. By day 200 only DNMT3B expression remained elevated.

Subjects: Sprague Dawley rats

Chemical: Bisphenol A

Low doses tested: 10.0 μg/kg bw/d

Route of administration: dissolved in tocopherol-stripped corn oil and delivered subcutanously

Exposure duration: post-natal day 1 – post-natal day 5 (comparable to human prenatal development from approximately day 1 of week 20 to day 1 of week 26)

Age of measurement: post-natal days 10, 90 and 200
